Off-the-Shelf CAR-T Achieves First Durable Remissions in Solid Tumors
Allogene's ALLO-316 just became the first off-the-shelf CAR-T therapy to produce durable remissions in metastatic solid tumors, with some patients cancer-free for over 18 months after a single infusion. The Phase 1 data, published in the Journal of Clinical Oncology, showed a 31% response rate in patients with high CD70-expressing kidney cancer who had already failed a median of four prior treatments. For a field that spent a decade failing to crack solid tumors with cell therapy, this is a genuine proof-of-concept moment.
Why it matters: If donor-derived CAR-T cells can achieve lasting responses in any solid tumor, it validates the entire engineering platform and opens the door to expanding cell therapy beyond blood cancers, where the vast majority of cancer deaths actually occur.
