

Eli Lilly is pouring $750 million into a manufacturing partnership with Resilience to expand KwikPen production for its GLP-1 blockbusters. In the obesity drug wars, the real bottleneck isn't science; it's making enough pens fast enough.
You can have the best-selling obesity drug on the planet. You can have insurance companies lining up to cover it. You can have patients begging their doctors for prescriptions. But none of that matters if you can't make enough of the stuff.
That's exactly the problem Eli Lilly is trying to solve. The company just dropped $750 million into a manufacturing partnership with Resilience, a contract manufacturer based in Ohio, to crank out more KwikPen devices for its blockbuster diabetes and obesity drugs, Mounjaro and Zepbound. The expansion will add at least 400 jobs and is expected to hit full operations by early 2027.
This isn't Lilly's first date with Resilience. The two companies started working together in 2023, and that partnership has already produced more than 150 million doses for U.S. patients. Now they're scaling the relationship from "seeing each other" to "moving in together."
And it tells you something important about where the GLP-1 race is really being won.
When people discuss the GLP-1 market, they usually focus on clinical data, insurance coverage, and pricing. All important. But the boring, unsexy constraint that has plagued both Lilly and Novo Nordisk for years is manufacturing.
Specifically, the bottleneck is something called fill-finish: the sterile process of putting the active drug into pens and assembling the final device. Think of it like this: you can brew all the beer you want, but if you don't have enough bottles and a bottling line, nobody's drinking.
The formal U.S. shortages for both tirzepatide (Lilly's drug) and semaglutide (Novo's) were resolved by early 2025. But "resolved" is doing a lot of heavy lifting in that sentence. National shortage designations ended because manufacturers convinced the FDA they could meet projected demand. Localized disruptions, though, can still pop up whenever demand spikes or a quality hiccup slows a production line.
The uncomfortable truth: most of the major new manufacturing capacity won't be fully operational until . That leaves a window where supply is tight, demand keeps climbing, and every additional pen you can produce is essentially printing money.

Lilly is not short on construction projects. The company's total U.S. manufacturing commitments since 2020 exceed $50 billion. That's not a typo. Fifty. Billion. Dollars.
A $9 billion site in Lebanon, Indiana is expected to start making medicines by the end of 2026. A $4.5 billion "Medicine Foundry" nearby opens in late 2027. There's a $3 billion Wisconsin expansion. And in February 2025, Lilly announced plans for four entirely new U.S. manufacturing sites, with locations to be revealed throughout 2025.
So why also spend $750 million with Resilience? Speed.
Building a pharmaceutical factory from scratch takes years. You need construction, regulatory inspections, validation runs, and FDA sign-off before a single commercial dose leaves the building. Resilience already has an operating facility in the Cincinnati area with an established workforce. Expanding an existing site is like adding a room to your house; building a new factory is like buying land and starting from the foundation.
By early 2027, this Resilience expansion could be churning out KwikPens while some of Lilly's owned sites are still in the validation phase. In a market where every quarter of production matters, that head start is enormously valuable.
Resilience is a fascinating company in its own right. Founded in 2020 at the height of COVID, it was born from the idea that U.S. biomanufacturing was dangerously fragile. The company raised over $2.4 billion in venture funding (led by ARCH Venture Partners and 8VC) and went on an acquisition spree, snapping up multiple manufacturing sites.
The company serves 65-plus clients and produces over 100 million commercial drug product units annually.
But Resilience hasn't had a perfectly smooth ride. Recent reporting indicates the company is streamlining its footprint, winding down six facilities while seeking additional debt financing. It's a pivot from "buy everything in sight" to "focus on what's working." The Lilly deal, one of the largest single investments in the company's history, validates that more focused strategy.
This is the part where we zoom out.
The GLP-1 market is expected to be one of the largest drug categories in history. Lilly and Novo Nordisk are the two dominant players, and both are spending staggering sums to expand production. Novo has committed $6 billion to capacity expansion; Lilly's commitment dwarfs that at roughly $9 billion earmarked specifically for GLP-1-related capacity, on top of its broader manufacturing investments.
But neither company can flip a switch. New manufacturing lines take years to build and validate. That means the company that can produce more doses sooner captures more market share, earns more revenue, and builds stronger relationships with insurers and pharmacy benefit managers.
Manufacturing capacity is becoming a moat, not just a cost center. It's the difference between a restaurant with a two-hour wait and one that seats you immediately. Both might have great food, but customers go where they can actually get a table.
Lilly's $750 million Resilience investment is a bet that in the GLP-1 wars, the winner won't just be the company with the best molecule. It'll be the one that can get those molecules into patients' hands fastest. And with additional drugs in the pipeline (including the oral GLP-1 candidate orforglipron and the triple-agonist retatrutide), Lilly is building manufacturing infrastructure not just for today's blockbusters, but for a whole generation of obesity and metabolic medicines.
Wall Street tends to get excited about clinical trial results and FDA approvals. Those are the glamorous moments. But increasingly, the real competitive advantage in pharma looks less like a data readout and more like a factory floor.
Lilly is spending like a company that believes demand for GLP-1 drugs will only grow, and that supply, not science, will be the limiting factor. At $750 million, the Resilience deal is a rounding error in Lilly's broader $50 billion manufacturing build-out. But it's a strategically important rounding error: near-term capacity that bridges the gap until the mega-factories come online.
