Off-the-Shelf CAR-T Cracks the Solid Tumor Problem for the First Time

Every allogeneic CAR-T therapy ever tested against solid tumors has failed. Allogene just broke that streak. Its donor-derived ALLO-316, published in the Journal of Clinical Oncology, produced durable remissions in advanced kidney cancer patients who'd already exhausted other options. The longest response has been ticking past 18 months with no signs of stopping. In patients with high CD70 expression, nearly a third responded to a single infusion, and median overall survival in that subgroup hasn't been reached. Zero cases of graft-versus-host disease were reported.

Why it matters: This is the first proof that off-the-shelf CAR-T can deliver lasting responses in solid tumors, potentially reshaping the cell therapy manufacturing paradigm and opening the door to scalable, lower-cost cancer immunotherapy beyond blood cancers.
